Make sure there is a Serial Port available on the PC and that there are no other applications using it.

A common cause of this problem is "Hotlink" software for hand-held computers, (i.e. Palm Desktop) that hold a serial port "Open" all the time. Shut down applications like this before trying to run the diagnostic system.
